This is a great reagent for detecting the protein content of samples. I used it for a vomit and diarrhea high school lab activity and it worked wonders!

Disposal

This disposal method is published for your convenience. You MUST have checked with your federal, state, and local regulations before using this method and these methods are only applicable for small laboratory sized quantities.

Use a lab apron, chemical resistant gloves and goggles before following the below method to dispose of hazardous waste.

 1. The concentration of the base should be 1 M or less. Dilute it to this concentration if it is more concentrated. Use a large beaker and place it in a fume hood.
  a. When dissolving a base, heat may be evolved. Ice can be used to help control the heat.
  b. Work with a small amount of base at first, and then scale up.
 2. Slowly add dilute hydrochloric acid to the base until the mixture is neutralized to about a pH of 7.
 3. Pour the final solution down the drain with at least a 20-fold excess of water.

Solution Prep

To prepare Biuret's Reagent Solution, add 2.3 g of copper(II) sulfate pentahydrate to 225 ml of DI water. In a second beaker, add 775 ml of DI water. Place this beaker in an ice water bath. Next, add 308 g of sodium hydroxide to the 775 ml of DI water. Mix the sodium hydroxide solution with the copper sulfate solution. The solution should be blue.
