This kit demonstrates the effect of pH on enzyme activity using the enzymes invertase and amylase. The pH affects the activity of enzymes in two ways: it affects the charges on groups in the enzyme’s three-dimensional active site, and it alters the charges that hold the enzyme molecule together.

Altering the charges in the active site can greatly change enzyme activity, but these changes are usually reversible. When an enzyme molecule is denatured by pH changes, the effects are usually not reversible, and the enzyme becomes permanently inactive. Enzyme activity is measured using DNS reduction to ANS, which may be assessed visually or by spectrophotometer.

Ordering information: Reply card must be returned to obtain invertase and amylase enzymes. This activity includes pH paper, amylase solution, invertase solution, iodine solution, starch, buffer solutions, sodium hydroxide, hydrochloric acid, 3.5 dinitrosalicylic acid (DNS) solution, and sucrose. A hot-water bath or hot plate and lab glassware including beakers, test tubes, and graduated cylinders are needed but not included. Use of a spectrophotometer or colorimeter is optional (absorption 540 nm).
