Use PCR Based Testing for Contaminants

This unique activity offers an opportunity to practice technique and analyze lab results for an environmental PCR-based experiment. You can use the results of the experiment as a predictor of water pollution due to sewage contamination. Students will PCR amplify targeted bacterial DNA to detect contaminating bacteria early in the pollution cycle. Bacterial strains are provided for making dilutions, to determine the number of bacterial cells that are required to obtain a successful PCR result. PCR products are then analyzed by agarose gel electrophoresis. After completing the first experiment, students can test the quality of waterways in their local environment using the same process and materials. Comes with materials for 10 setups, including methylene blue and EtBr stains, and a teacher’s guide with complete instructions, background information, and study questions. A horizontal electrophoresis chamber, power supply, light box, variable volume micropipet, centrifuge, and thermal cycler, are all required and available separately.
